Synchrotron SAXS
data from solutions of
MsbA in stealth nanodisc (SAXS)
in
30 mM Tris, 150 mM NaCl, pH 7.5
were collected
on the
EMBL P12 beam line
at the PETRA III storage ring
(DESY; Hamburg, Germany)
using a Pilatus 2M detector
at a sample-detector distance of 3.3 m and
at a wavelength of λ = 0.124 nm
(I(s) vs s, where s = 4πsinθ/λ, and 2θ is the scattering angle).
Solute concentrations ranging between 1 and 2.7 mg/ml were measured
at 10°C.
20 successive
0.050 second frames were collected.
The data were normalized to the intensity of the transmitted beam and radially averaged; the scattering of the solvent-blank was subtracted.
MsbA ABC transporter protein encapsulated in a partially deuterated "stealth-nanodisc": (sND). Disc components (MSP1D1 scaffold protein - 75% deuterated, and POPC lipid - heads/tails 78% and 92% deuterated). The expected molecular weight for this protein-lipid complex should correspond to that of the MsbA protein (~132 kDa) + 2 x MSP1D1 (2 x 24.7 kDa) + the contribution of the POPC lipids (760 Da per monomer).
