Synchrotron SAXS data from solutions of the M domain of P. falciparum Heat shock protein 90 (PfHsp90) in 25 mM Tris-HCl, 100 mM KCl, 1 mM β-mercaptoethanol, 1 mM EDTA, pH 7.5 were collected on the SAXS1 Beamline beam line at the Brazilian Synchrotron Light Laboratory (Campinas, Brazil) using a Pilatus 300K detector at a sample-detector distance of 1 m and at a wavelength of λ = 0.1448 nm (I(s) vs s, where s = 4πsinθ/λ, and 2θ is the scattering angle). One solute concentration of 3.10 mg/ml was measured at 20°C. The data were normalized to the intensity of the transmitted beam and radially averaged; the scattering of the solvent-blank was subtracted.
Samples were measured for 6 x 10 seconds , 1 x 100 seconds and 1 x 300 second frames. The PfHsp90 sample concentrations were 1.2, 1.9 and 3.1 mg/mL. Data analysis was performed using the ATSAS 2.7.2 program. Ab initio modeling was performed using the DAMMIN program. Average of DA models was done by the DAMAVER package. Further refinement was done using the DAMMIN program.
