Synchrotron SAXS data from solutions of full-length human DPCD in 25 mM Tris-HCl, 200 mM NaCl, pH 7.5 were collected on the 16-ID (LiX) beam line at the National Synchrotron Light Source II (NSLS-II; Upton, NY, USA) using a Pilatus / 1M SAXS detector detector at a sample-detector distance of 2.3 m and at a wavelength of λ = 0.0819 nm (I(s) vs s, where s = 4πsinθ/λ, and 2θ is the scattering angle). Solute concentrations ranging between 0.2 and 3.6 mg/ml were measured at 20°C. 10 successive 1 second frames were collected. The data were normalized to the intensity of the transmitted beam and radially averaged; the scattering of the solvent-blank was subtracted. The data from the concentration series were extrapolated to infinite dilution.
DPCD data were collected from samples prepared at 0.23 mg/mL, 0.45 mg/mL, 0.9 mg/mL, 1.8 mg/mL, and 3.6 mg/mL in 25 mM Tris-HCl (pH 7.5), 200 mM NaCl. A single curve extrapolated to infinite dilution was obtained using Almerge in the ATSAS 2.7.2 package. Recombinant human DPCD was heterologously expressed in Escherichia coli. |
