The complete form of the protein of interest is a fusion protein between the Escherichia coli Maltose-Binding Protein (MBP) (Uniprot: P0AEX9) and the Mycobacterium tuberculsosis MabR (Rv2242) (Uniprot: P9WPH5). The MBP used is a variant with an additional linker and point mutations (D108A, K109A, E198A, N199A, K265A). The molecule described here is the MabR part of the fusion protein and contains an additional non-native polyhistidine tag at the N-terminus (HHHHHH).The fusion protein adopts three forms in solution: tetramer, dimer and monomer. Then, in SEC-SAXS three frames are averaged and analyzed for the three different peaks.
